Arhgef2 (Rho/Rac Guanine Nucleotide Exchange Factor 2), also called GEF-H1 is a Rho-GTPase GEF and a microtubule binding protein. Microtubule depolymerization causes detachment of Arhgef2 from microtubule cytoskeleton, released Arhgef2 activate Rho-GTPases, which modulate actin cytoskeleton. CDKL5 phosphorylates mouse Arhgef2 at Serine 122, in vitro, this site is conserved in human and rats.
