The gene encoding Bcl-3 was first identified as the gene altered in some cases of chronic B cell lymphocytic leukemia and was later found to belong to the IκB subfamily of inhibitors of NFκB transcription factors. Bcl-3 does not have DNA-binding activity, but it interacts with NFκB1 and NFκB2 which, when bound to DNA, repress gene transcription. Therefore, Bcl-3 may promote gene expression that is repressed by members of the NFκB family. Bcl-3 RNA is highly expressed in spleen and liver with lower levels of expression in heart, skeletal muscle, lung, kidney, and testes.
