SARS-CoV-2 (Severe Acute Respiratory Syndrome Coronavirus 2) also known as 2019 nCoV (2019 Novel Coronavirus) is an enveloped, single, and positive-stranded RNA virus. The virus encodes four major structural proteins that have played important roles in causing the infection: Spike protein (S protein), nucleocapsid protein (N protein), membrane protein (M protein), and small envelope protein (E protein).
S1 sub-unit binds via its RBD (Receptor Binding Domain) to the cellular receptor angiotensin-converting enzyme 2 (ACE2) and active S2 sub-unit fuses to the viral membranes with the host cell membrane. The spike protein is reported to induce the neutralizing-antibody and T-cell responses, as well as protective immunity. The CoV spike (S) glycoprotein therefore represent the key target for vaccines, therapeutic antibodies, and diagnostics.
