Peroxidase labelling has the advantage of being sensitive and permanent.
Peroxidase substrates are varied: precipitating substrates for western blot or immuno-histochemistry (DAB, AEC, 4CN, TMB), soluble substrates for ELISA (TMB, OPD, ABTS) and chemiluminescent substrates (luminol).
However, peroxidase labelling has some disadvantages: Peroxidase is rendered inactivated by oxygen, hypochloric acid and sodium azide.
It is therefore advisable to use highly purified water.
The presence of endogenous peroxidase in certain tissues like macrophage, red blood cells, bone marrow cells and plants could increase the background.

Antibodies are particularly suitable for use in the most sensitive procedures (ELISA, immuno-histochemistry, immuno-cytology and western blot).
Their quality is guaranteed because a very stringent purification protocol is followed where non-specific and specific purifications alternate (affinity chromatography).
Antibodies undergo a routine inspection of the cross reactions between target species.
If necessary, the total elimination of cross-reactions can require an adsorption by affinity chromatography against the antigen whose recognition is not desired.
Their quality is guaranteed because a very stringent purification protocol is followed where non-specific and specific purifications alternate (affinity chromatography).
