Fluorescein labelling is carried out to obtain a good signal while at the same time minimising the background.
The F/P molar ratio is set between 3 and 7.
FITC has the advantage of being detectible under a microscope.
It can be easily used for carrying out multiple labelling in histochemistry or cyto-chemistry.
However, FITC may rapidly lose its fluorescence.
Maximum absorption: 495 nm.
Maximum emission: 520 nm.
Colour: green yellow.
Applications
Antibodies are particularly suitable for use in the most sensitive procedures (ELISA, immuno-histochemistry, immuno-cytology and western blot).
Their quality is guaranteed because a very stringent purification protocol is followed where non-specific and specific purifications alternate (affinity chromatography).
Antibodies undergo a routine inspection of the cross reactions between target species.
If necessary, the total elimination of cross-reactions can require an adsorption by affinity chromatography against the antigen whose recognition is not desired.
