Covalab are experts in the use of synthetic peptide antigens to target specific antigenic epitopes. Raising antisera to immunogenic peptides is the simplest method of obtaining antibodies against non-isolated proteins, for example, against putative protein sequences derived from DNA sequence information.

This approach is also useful when the isolation of the antigen is difficult or time consuming, or when the antigen is a member of a large protein family. To produce antibodes against synthetic peptides we offer two levels of service.

To undertake the design of a peptide, we ask for the following information: |
- Name of the protein
- Sequence (FASTA format) by email or computer compatible support (Disk, CD-ROM) if you project is confidential
- Regions of the protein in which you are interested (N-ter or C-ter)
- Specificity required by comparison to other proteins
- List of homologous proteins

Once all this information is available, an analysis of the different parameters such as hydrophobicity, antigenicity, antigenicity, accessiblility with solvent, presence of transmembrane regions and a blast of sequences will be made. |

You can use 1, 2, or 3 peptides for your antibody development. However this possibility depends on the results of the peptide design. All peptides are produced with Fmoc strategy on solid phase and analyzed by HPLC to ensure purity and by mass spectrometry to dertermine the molecular mass.

The quantity to be synthesised depends on the service chosen :

- Standard Service : peptide 15 AA, 5-10 mg at 70% of purity, no modification
- Package Service : peptides 15 AA, 20-25 mg at 70% of purity, no modification (exception - Quantity synthesised for producing antipeptide antibodies in goats : 50 mg)

Nevertheless, you can choose to synthetize peptides with other quantities, purities, sequence lenght... |

Usually we conjugate the peptide to a carrier protein via a cysteine residue. If the amino acid sequence of the peptide does not contain a natural cysteine, we add a cysteine to Nter or Cter sequence of the peptide. |

Covalab enables you to select the rabbits best suited for your work, by sending you pre-immune bleedings of animals. Only supplementary sampling will be billed.
(Example : if two rabbits to be immunized are ordered, 5 bleeding will be sent to you, but only 3 will be billed.

Each sampl will be tested for anti-peptide antibody using the CovAbtest®. It enables the serum's immunoreactivity to be tested on antigens (peptide(s) injected), previously coupled covalently to the walls of 96-microwell plates. These elements and the results obtained in your lab enable you to take the right decision. |
