CUSTOM POLYCLONAL ANTIBODY
ANTIBODY AGAINST POST-TRANSLATIONAL MODIFICATION

PEPTIDE SYNTHESIS
2 peptides will be synthesized to perform a project:
- 1 modified peptide: <16 AA, 20-25 mg at >70% of purity
- 1 non-modified peptide: <16 AA, 5-10 mg at >70% of purity
Nevertheless, you can choose to synthesize peptides with other quantities, purities, sequence length or adding modifications.
CONJUGATION
Normally, peptide is conjugated to a carrier protein via a cysteine residue. If the amino acid sequence of the peptide does not contain a natural cysteine, we add a cysteine to Nter or Cter sequence of the peptide.
- Only the modified peptide is conjugated to a carrier protein
- Carrier proteins available: Keyhole Limpet Hemocyanin (KLH), Bovine Serum Albumin (BSA) or Ovalbumin (OVA)
IMMUNIZATION
Two rabbits will be immunized with the modified peptide following our 88 Days protocol.
According to the results, protocol may be changed after discussion with customer.
ELISA
Each test bleeds will be tested against each peptide by
ELISA.
These ELISA test will allow you to follow the evolution of the
immuno-reactivity of the rabbits during the immunization process.
IMMUNO-AFFINITY PURIFICATION
Covalab performed a specific immune-affinity purification that allows obtaining specific antibody against site specific modification of protein.
- 2 Immuno-Affinity columns will be prepared (1 with the modified and 1 with the non-modified peptide)
- Purification in two steps:
- Loading of 5mL of sera on the non-modified peptide agarose column
- Loading of the non-retain sera on modified peptide agarose column
