CUSTOM POLYCLONAL ANTIBODY

ANTIBODY AGAINST PEPTIDE

Using synthetic peptides as immunogens is the simplest way to produce antibodies against non-purified proteins or against putative protein sequences derived from DNA sequence information. This method is also useful when the isolation of the antigen is difficult and time consuming or if the antigen is a member of a large protein family. In order to provide best quality antibodies for customer, discussion with customer will be done to understand their needs and to advice for the best approach. During the project, a complete follow by Covalab will help customer to take right decision.

A Package Service including ELISA titration and immunopurification can be providing by Covalab.

PEPTIDE DESIGN

To perform the peptide design, the following information will be required:

  • Protein name
  • Sequence (FASTA format)
  • Domains of interest (N-ter or C-ter, cytoplasmic or extracellular)
  • Specificity required by comparison to other proteins
  • List of homologous proteins
  • Application (WB, IHC, IP...)

Based on the supplied information on the protein, an analysis of the different parameters such as hydrophobicity, antigenicity, accessiblility with solvent, post-translational modifications, presence of transmembrane regions and a blast of sequences will be performed to design the most relevant peptide.
A list of designed peptides will be send to customer.

PEPTIDE SYNTHESIS

All peptides are produced with Fmoc strategy on solid phase and analysed by HPLC to ensure purity and by mass spectrometry to determine the molecular mass.

Quantity of peptide to be synthesised will depend on the service chosen:

  • Standard Service: peptide <16 AA, 5-10 mg at >70% of purity
  • Package Service: peptides <16 AA, 20-25 mg at >70% of purity

CONJUGATION

Normally, peptide is conjugated to a carrier protein via a cysteine residue. If the amino acid sequence of the peptide does not contain a natural cysteine, we add a cysteine to Nter or Cter sequence of the peptide.

Carrier proteins available: Keyhole Limpet Hemocyanin (KLH), Bovine Serum Albumin (BSA), Ovalbumin (OVA)...

Quantity of peptide conjugated: 3-5mg or more in case of using goat or sheep as animal host.

IMMUNISATION

According to the preliminary results, customer's protocol can be adapted after consultation. At the end of the standard protocol, the final bleed will be never proceeding without customer's confirmation.
